Improvement of Safflower (Carthamus tinctorius L.) for Salinity Tolerance under in vitro Condition

Aim: The primary aim of the present study was to screen salt tolerant calli and optimization in-vitro regeneration protocol from selected calli.
 Methodology: cotyledonary leaf explants sterilized by using 1% Bavistin, 0.1% Mercuric chloride 70% ethanol followed washing with distilled water. Sterilized were cultured on Murashige Skoog (MS) medium supplemented different concentration NaCl (i.e. 0, 50mM, 100mM, 200mM, 300 mM 400 mM) check salinity tolerance ability PKV pink genotype. In vitro screening callus studied morphological characters like colour texture callus, growth percentage, relative water contained, cell survivility proline content saline stress unstress confirm pressure regenerate Pink safflower genotype.
 Results: MS 150 showed 50% survival calli, whereas no obtained in high NaCl. Moreover, biochemical assay estimation done for its confirmation calli. accumulation found be highest grown media as compared control. Also observation i.e. viability under select line Pink. shoots failed produce rooting standardized medium. So, approach higher auxin shock grafting experiment attempted overcome problem. Higher but satisfactory shoot development Conclusion: abiotic plants better understanding physiological changes conditions.
